In this film, the historic conquistador Lope de Aguirre and his fictitious brother-in-arms Francisco Lopez de Heredia are major characters. These conquistadors attempted to steal the magical Tears of the Moon, resulting in them being cursed with immortality by the Indigenous Puka Michuna tribe. By 1916, Francisco became the Jungle Navigation Company, "Skipper" Frank Wolff while Lope was driven mad and recruited by German prince, Joachim of Prussia.

The conquistador helm of Lope de Aguirre is hung within the Jungle Navigation Company's boathouse in the Lost Delta of India. Additionally, a Spanish map and clothing of Frank Wolff are hung in both this boathouse and the company's Amazon River Base. During Indiana Jones Summer of Hidden Mysteries, Trader Sam could be seen with a tablet depicting the Crystal Skull of Akator which was stolen by conquistador Francisco de Orellana in 1546.
The depictions of Indigenous South American characters such as Trader Sam and Chief Nah-mee is also influenced by stereotypes created by the Spanish Empire of the Jivaroan peoples. The Jivaroan spent much of their history successfully fighting off the Spanish Empire resulting in the Empire spreading propaganda about them being dim-witted and savages.
Skipper Canteen[]
In the Skipper Canteen's library is a book titled, "The Fountain of Youth" by Diego. This seemingly alludes to Diego Colon, the son of colonist Cristobal Colon who aided the Spanish Empire in the conquering North America before they arrested him for various atrocities against the Indigenous population. Diego Colon himself was a viceroy for the Spanish empire while his murderous father was named an honorary member of the Society of Explorers and Adventurers.

In this film, the Spanish Empire attempted to compete with the British Empire and pirate Blackbeard to find the Fountain of Youth. Ultimately, the Spaniards destroyed the fountain as they believed it to be an affront against their Catholic God.
